Have you ever looked at this area before? It is normal for some people to have patches of darker skin there. I would keep an eye on it. If it changes shape or spreads, then see a doctor. Other wise I wouldn't worry.
As for the anal sex, it's important to use lots if lube and take things really slowly. Particularly if it's your first time.
