What I would do is to get is some liquid antiseptic (TCP if you are in the UK) and carefully place a tiny drop of this on it twice a day - it may sting initially but it should help with the healing process. It is a skin irritation of some kind - it could be you are getting a build-up of fluid of some kind and this is exploiting a slight imperfection in the skin, or the skin has become damaged in some way in this area.
As I say, normal healing will not occur as the skin there is not exposed to the air.
Try regularly exposing the entire area and wash with water only, then you need to make sure the entire head, foreskin and upper shaft of the penis are completely dry. Take time to carefully pat dry with a clean towel this particular area so as not to cause any damage.
